(AP) ? A Syrian activist group reported Monday that 144 people have been killed across the country, scores of them in the embattled opposition stronghold of Homs by security forces as they tried to flee. A team from the Syrian arm of the Red Cross delivered aid to one of the city's most dangerous neighborhoods after days of trying to reach the area.

The activist group did not say whether all 144 died on Monday or were killed over the past few days. Many of the casualties were believed to be from the rebel-controlled Baba Amr neighborhood of Homs, which the Syrian Arab Red Crescent entered late Monday. Also in the neighborhood are two wounded foreign journalists along with the bodies of two of their colleagues who were killed last week.

European and American diplomats and aid workers have been trying desperately to find a way to evacuate them, but Red Cross spokeswoman Carla Haddad said late Monday that the Red Crescent had not managed to get them out. She did not know whether the group had stopped trying for the evening.

Homs has emerged as the center of the 11-month-old uprising seeking to oust authoritarian President Bashar Assad and has borne the brunt of his regime's bloody crackdown on dissent. Parts of the city have been surrounded for weeks, making it impossible for rescue workers to reach the wounded and for families to bring their dead and injured to the hospital.

Reports by numerous activists that more than 60 bodies were brought to the hospital, all of whom appeared to have died in one incident, reflect the spreading carnage.

The high death toll reported by the Local Coordination Committees activist group is sure to add to the growing international pressure on Assad to give up power. But so far, his regime has shown no signs that it is ready to leave peacefully.

Syrian officials announced the results of a referendum on a new constitution held Sunday that Syrian authorities lauded as a step toward political reform.

The referendum allows at least in theory for opening the country's political system. It approves a new constitution, which allows for a multiparty system in Syria, which has been ruled by the Baath party since it took power in a coup in 1963. Assad's father, Hafez, took power in another coup in 1970.

It also imposes a limit of two seven-year terms on the president, meaning Assad could remain legally in power through 2028.

The U.S. and its allies dismissed the vote as a "farce" meant to justify the regime's bloody crackdown on dissent. Syria's main opposition groups boycotted the vote, and violence elsewhere prevented polling.

Syrian state TV said 89 percent of eligible voters approved the new document, while nine percent rejected it. It put turnout at 57 percent of Syria's 14.9 million eligible voters.

Representatives of more than 60 countries met in Tunisia last week to forge a unified strategy to push Assad from power and began planning a civilian peacekeeping mission to deploy after the regime falls. On Monday, the European Union imposed new sanctions.

Syria has been able to count on allies China and Russia to protect it from condemnation by the U.N. Security Council. Both staunchly opposed any interference in Syria's affairs.

Russian Prime Minister Vladimir Putin warned Monday against military intervention in Syria and blasted the West, saying it had backed the Arab Spring to advance its interests in the region.

In Damascus, a Syrian official accused the West of trying to destabilize the country for its own gain and warned that militarizing the opposition is a big mistake that will backfire.

Foreign Ministry spokesman Jihad Makdissi told The Associated Press in a rare interview that dialogue among all parties is the only way to end the conflict.

"The West took advantage of the awakening of the Syrian street. Instead of helping Syria to overcome this painful crisis, they are using this to hit the stability of Syria for other geopolitical reasons," he said.

Activists groups said Monday that the death toll for 11 months of unrest has now surpassed 8,000 people.

The diplomatic pressure on Syria comes as the humanitarian and security situation on the ground is collapsing. The Local Coordination Committees said 64 of those reported dead on Monday were fleeing shelling in the Baba Amr neighborhood of Homs when security forces stopped them at a checkpoint in the city's Abil area and killed them.

Another activist in Baba Amr, reached via Skype, said a security officer who had seen a report on the incident leaked its contents to activists, with whom he sympathizes.

The activist, Abu Mohammed Ibrahim, said the group left the neighborhood Sunday evening and were stopped at a checkpoint, where security forces loaded them onto four busses. A while later, the buses stopped and the soldiers started killing passengers. Locals found the bodies dumped in two places outside of town on Monday, the activist said.

A second activist group, the Britain-based Syrian Observatory for Human Rights, said 68 bodies were brought to the hospital in Homs and that they were found between the villages of Ram al-Anz and al-Ajriyeh.

But the group's head, Rami Abdul-Rahman, said no one had identified the dead or knew where they were from.

"We know they are from Homs, but we don't know anything for sure about who they are or how they died," he said.

None of the death tolls could be independently verified.

Some of the worst fighting in Syria's nearly one-year-long conflict has come in Homs, where residents have been bombarded by Syrian government forces for nearly four weeks.

Two western journalists were killed in government shelling in Homs last week, and two other journalists injured.

Poland said Monday its diplomats are working with U.S., British and French authorities to evacuate the two reporters ? Edith Bouvier of France and Paul Conroy of Britain ? as well as the bodies of American Marie Colvin and Frenchman Remi Ochlik, who were killed in the same attack.

French President Nicolas Sarkozy accused Syrian authorities of assassinating Colvin and Ochlik by targeting a makeshift media center where they were killed.

"When the Syrian army fires shells several times on a building that they perfectly well knew was a press center ... it's an assassination," he said during an interview with RTL.

A: Don't be so sure. There are many programs out there that can help reduce your loan payments. It comes down to four factors: math, persistence, luck and realistic expectations. By math, I mean that your income, bills, home value and mortgage must fit into one of your lender?s programs. As for persistence, many people get turned down two or three times before they get approved, so if you stop trying you?ll never get the relief you?re seeking. There is an element of luck involved in all of this. Did you randomly get a bank representative who will go the extra mile to help you, or the one who is in the middle of a divorce and hates his job? Finally, it?s important to have realistic expectations. I have seen many people turn down a terrific loan modification offer from the bank to wait it out for some magical solution that?s never going to happen.

You need to get an idea of what type of relief that you may qualify for, and see if that relief will work for you. For example, if you earn $700 a month in Social Security as your only form of income and you are trying to modify a $300,000 mortgage, it is highly unlikely that you are going to be offered a solution because the math just doesn?t work. If you earn $200,000 a year and are trying to modify your $190,000 mortgage, it?s doubtful that your lender is going to see things your way.

Q: Our mortgage is underwater and we would like to move out of the immediate area. I need some good advice. I'm reluctant to just pick up the phone an call anyone because I don't want to become a victim of a scam. Do I call a law firm? A real estate agent? A loan modification company? Who? ? Hal

A: Call a lawyer experienced in dealing with struggling homeowners. He or she will help you figure out a strategy that may involve a short sale or a loan modification. Each person?s situation is unique and each situation should be dealt with individually. Do some Internet research to find out about lawyers in your area and then choose two or three to interview. Ask questions such as: How many cases have you handled similar to mine? Do you have any special training or certifications related to real estate law? You should also take to the appointment appropriate paperwork about your financial situation and your house. Bring proof of income for all wage earners, two years of tax returns, a recent mortgage statement, your tax bill, your insurance bill and any other paperwork that you think is pertinent to your individual case. Think about your goals. Attorneys are, by training, goal-oriented people, so tell the attorney about your goals so he or she can craft a custom plan that works for you. Bring reasonable expectations and a willingness to listen.

A weakness in cancer's defense system exploited

Researchers at the EPFL have identified an important mechanism that could lead to the design of more effective cancer vaccines. Their discovery of a new-found role of the lymphatic system in tumour growth shows how tumours evade detection by using a patient's own immune system.

Tumour cells present antigens or protein markers on their surfaces which make them identifiable to the host immune system. In the last decade, cancer vaccines have been designed that work by exposing the patient's immune cells to tumour-associated antigens and so priming them to kill cells that present those antigens. These have caused much excitement, not least because by acting so specifically on cancer cells, they could potentially eliminate the unpleasant side effects of chemo- and radiotherapy.

However, clinical trials of such vaccines have had a very low success rate to date, mainly because tumours have various mechanisms for evading detection by immune cells, even when those immune cells called T cells have been primed to seek them out. Those mechanisms are, in general, poorly understood. But in a paper to be published this week in Cell Reports, the laboratories of Melody Swartz at EPFL and Stphanie Hugues at UNIGE provide a key insight into one of them. They describe for the first time how, like soldiers protecting a fort, lymph vessels surrounding a tumour ward off T cell attack.

Plenty of research has shown that tumours can induce the growth of lymph vessels in their vicinity, and that this growth is correlated with metastasis and poor prognosis. It was assumed that these lymph vessels simply provided an escape route for cancer cells, transporting them to distant sites. In the new study, led by postdoc Amanda Lund, the researchers show that lymph vessels actually suppress the immune response, deleting the attacking T cells or leaving them "functionally exhausted" by the time they reach the tumour.

They studied a type of tumour that expresses large amounts of VEGF-C, a molecule that is naturally expressed in humans and that stimulates lymphatic growth. Having engineered the tumour cells to express a foreign antigen, they compared the efficacy of a vaccine designed to prime T cells to kill cells carrying that antigen, either when VEGF-C was present or when its activity was blocked . With VEGF-C suppressed, the vaccine's efficacy increased and tumour growth slowed fourfold.

A weakness in cancer's defense system exploited

The researchers went on to show that the endothelial cells which line lymph vessels "scavenge" tumour-specific antigens and present them to the tumour-specific T cells in a suppressive manner. This, in turn, promotes the local deletion of those T cells. According to Prof Swartz, that means that first targeting the lymph vessels associated with a tumour could, in theory, significantly increase the efficacy of existing cancer vaccines. "It would be like removing the soldiers from around the fort before sending in your opposing army," she says. "If you disable the lymph vessels' suppressive functions, our data suggest that tumour-killing T cells would do their job a lot more effectively." Future clinical trials are needed to put that theory to the test.

Opening GOP debate, Romney pounces on Santorum

MESA, Ariz. (AP) ? Republicans Mitt Romney and Rick Santorum are sparring in what is likely to be the final debate before the next slate of states has nominating contests, trading barbs on Washington spending and borrowing.

Romney on Wednesday said government doubled in size while Santorum served in the House and Senate. Romney notes that Santorum voted to increase the nation's borrowing limit five times without corresponding budget cuts, as well as voting to send tax dollars to Planned Parenthood.

By contrast, Romney says he balanced budgets during his four years as Massachusetts governor and at the helm of the Olympic Games.

Santorum is not letting Romney have a pass on it. He says that Romney supported last year's vote to increase the nation's borrowing limit to avoid a government default.

The home stretch | let's be moms

Chrissy is nearly 34 weeks pregnant. It seems like the girls will be here ANY day now. So, naturally, there?s a lot going on at our house!

After talking with Chrissy?s OB (who we see every week now), we decided to go ahead and schedule the C-section. March 28. Our babies? birthdays could be March 28. A little more than a month away. However, our OB and the high-risk doctors (who we see twice a week) agree, the girls will probably be here before then!? Each day brings an increased risk for complications and if there are complications that could endanger the babies or Chrissy, they will perform the C-section. We want them to stay in as long as possible so they are strong and healthy.

Even though we have a lot of ultrasounds done (pretty much twice a week now), we don?t get to see the girls faces very often. The doctors prescribe the ultrasounds to measure the fluid surrounding both babies, monitor heart rates and movement. This week, though, we had a rare and precious glimpse at their little faces.

We?re trying to kick gestational diabetes in the butt. Chrissy is doing a great job?avoids the sweets, carbs and really sticks to the healthy food diet. In other news? We had an amazing time at our baby shower last month. Our family did a wonderful job working together and organizing everything for a wonderful, memorable day. Everything was beautiful and thoughtful. And it was so great to catch up with our friends and family we?ve been neglecting all this time!

Researchers reveal role of protein mutation in Parkinson's disease

Purdue University researchers revealed how a mutation in a protein shuts down a protective function needed to prevent the death of neurons in Parkinson's disease, possibly opening the door to new drug strategies to treat the disorder.

Fred Regnier, the J.H. Law Distinguished Professor of Chemistry, and Jean-Christophe Rochet, an associate professor of medicinal chemistry and molecular pharmacology, led the team that discovered how the protein DJ-1, which plays a significant role in protecting neurons from damage, is shut down by a subtle mutation.

A substitution in one link of the chain of amino acids that makes up the protein renders it unable to be activated to protect neurons from the build up of protein "aggregates," or "clumps," that lead to cell death in those with Parkinson's disease.

"The saying that you are only as strong as your weakest link appears to hold true in the case of the chain of amino acids that make up a protein," Regnier said. "The magnitude of the effect of this subtle change is surprising. It can make the difference between having a disease and being healthy."

According to the Parkinson's Disease Foundation, an estimated 7 million to 10 million people worldwide are living with the disease, which is a neurodegenerative disorder that causes muscular rigidity, slowness of movement, poor balance and tremors. The death of neurons in a region of the brain called the substantia nigra cause the symptoms.

The findings of the Purdue-led study could potentially lead to new Parkinson's treatments, Rochet said.

"The current methods of treatment are to add back what the lost cells used to produce, similar to hormone replacement therapies," he said. "Understanding this error in a key protein could help researchers find a way to prevent cell death in the first place. Perhaps a compound could be found that could correct the problem and resurrect the protective function of the protein. Of course interventions would be needed in many places to treat the disease, but this could be one of several places to target for a potential treatment."

When functioning properly, DJ-1 appears to serve as a "chaperone" protein for the neural protein alpha-synuclein, escorting and protecting it as it performs its biological task. Without the help of DJ-1, alpha-synuclein can unfold and expose sticky surfaces that cause it to clump together with other proteins. These clumps are a component of the "Lewy bodies" and other protein deposits that build up in the neurons of Parkinson's disease patients and cause the cells to die, he said.

About 10 years ago it was discovered that people with familial, early-onset Parkinson's disease had a mutation in the gene that encodes DJ-1 that leads to a mutant form of the protein through a substitution in one of the protein's amino acids.

The Purdue-led team developed a new quantitative mass spectrometry approach to evaluate and compare the mutant and normal protein. They discovered that the substitution prevents DJ-1 from undergoing an important chemical reaction in which oxygen is added to a specific site on the protein. This addition of oxygen takes the protein into a two-oxygen form that facilitates its chaperone function.

It had been thought that the amino acid substitution led to an unfolding of the protein, but the team found that it instead slightly alters the structure of the active site pocket, preventing the addition of oxygen at that site.

In addition the team found that the attachment of too much oxygen or an oxygen atom linked at the wrong location also disabled the protein's protective abilities, Rochet said.

"The interaction of this protein with oxygen needs to be very precise," Rochet said. "We need just enough oxygen added at just the right site to activate the protective ability of the protein, but too much oxygen or oxygen added at the wrong location causes real problems."

Because the precise oxidation of the protein may play a significant role in preventing the development of Parkinson's disease, evaluation of the levels of oxidized DJ-1, non-oxidized DJ-1 and over-oxidized DJ-1 could be the starting point of a new diagnosis method, Regnier said.
